Methodological: In addition to conventional cytogenetic and molecular genetic procedures necessary for the manipulation and analysis of chromosomal DNA and RNA (PCR, RT-PCR, Southern blotting, DNA sequence analysis, RFLP analysis, etc) we routinely use fluorescent in situ hybridisation (FISH) and its many applications for our research. These applications include gene and genomic mapping on metaphase and interphase nuclei, whole chromosome painting, use of centromeric probes for chromosome enumeration, and comparative genomic hybridisation (CGH). We have recently extended these applications to include gene expression analysis by microarray. We also have an extensive background of experience using high-resolution expression microarrays, and also array comparative genomic hybridisation (array CGH) for assessment of structural variants including DNA copy number variations (CNVs) that are associated with predisposition, cause and progression of many human disease conditions, including cancer.
